CJC-1295 vs Ipamorelin: what the studies show

CJC-1295 and ipamorelin both push the body to release growth hormone, but they work in different ways. Neither has a human trial showing muscle, fat-loss or anti-aging benefits. CJC-1295 has hormone-level data in healthy adults, while ipamorelin's main efficacy trial found no significant benefit.[1][2][3]

What each one is

CJC-1295 is a synthetic copy of growth-hormone-releasing hormone (GHRH) with four amino-acid substitutions that help it resist breakdown. It makes the pituitary release growth hormone (GH), which raises IGF-1.[1]

There are two products under one name. The DAC form binds albumin and its effects last days. The non-DAC form is essentially uncharacterised in people, and all the controlled human data are for the DAC form.[1][2][4]

Ipamorelin is a five-amino-acid peptide that acts on the ghrelin receptor and also makes the pituitary release GH.[5][6][7][3]

Our sources don't cover a head-to-head study of the two.

What the human studies show

For CJC-1295, two randomised, double-blind, placebo-controlled studies in healthy adults aged 21 to 61 gave ascending doses under the skin. After one injection, mean GH rose 2- to 10-fold for 6 days or more. IGF-1 rose 1.5- to 3-fold for 9 to 11 days.[2]

The half-life was estimated at 5.8 to 8.1 days, and no serious adverse reactions were reported.[2]

In a separate non-randomised study, healthy men aged 20 to 40 were sampled before and one week after a single injection. Mean GH rose 46% and IGF-1 rose 45%.[8]

Those are hormone measurements, not proven benefits. FDA's reviewers said the human studies did not measure outcomes such as body composition.[4]

A phase 2 trial in HIV patients with visceral fat is listed as terminated, with no results in the record.[9]

For ipamorelin, the human record is also thin:

  • A 1999 dose-escalation study used 8 healthy men at each of 5 doses. The half-life was about 2 hours, and each dose was followed by a single episode of GH release. It did not test any health benefit.[10]
  • A phase 2 trial in 114 adults after bowel resection compared ipamorelin 0.03 mg/kg IV twice daily for up to 7 days with placebo. Median time to first tolerated meal was 25.3 hours on ipamorelin and 32.6 hours on placebo. That was not a significant difference (p=0.15).[3][11]
  • A larger dose-finding trial (enrollment 320) is registered as completed, but our records hold no results from it.[12]

Rat studies of bone and muscle have no matching human trial in our records.[5][13]

Safety signals and regulators

For CJC-1295, FDA described the 2006 healthy-adult studies this way. Injection site reactions occurred in about 70% of recipients in one study and in all recipients in the other. Heart rate rose after a single dose in another study.[4]

FDA also described anecdotal internet reports of a participant death in a phase 2 HIV trial of the DAC form. The physician said the most likely explanation was coronary artery disease without symptoms, and FDA said the data had not been published.[4]

FDA says long-term human safety is unknown and that antibody risk with long-term repeated use cannot be ruled out.[4]

For ipamorelin, adverse events in the bowel surgery trial were 87.5% on ipamorelin and 94.8% on placebo. The authors called it well tolerated.[3]

FDA states that a published study found serious adverse events, including death, when ipamorelin was given IV to improve gastric motility. We could not match this to a specific result in the trial abstract.[14][3]

On the regulatory side:

  • At its December 4, 2024 meeting, FDA's advisory committee voted 0 yes and 13 no on the CJC-1295 free base for the 503A bulks list.[15]
  • On 2024-10-29, the committee recorded zero yes votes, 12 no votes and 1 abstention on ipamorelin free base.[16]
  • FDA's category 2 page lists CJC-1295 under substances nominated but withdrawn, citing immune-reaction risk, increased heart rate, a vasodilatory reaction and limited clinical data.[14]
  • The same page lists ipamorelin acetate, for 503B outsourcing facilities, among bulk substances that may present significant safety risks for compounding.[14]
  • A warning letter dated 2020-04-01 to one pharmacy, Tailor Made Compounding LLC, listed CJC-1295 among bulk substances it compounded that were not eligible for 503A exemptions.[17]
  • FDA has also cited a pharmacy for compounding with ipamorelin.[17]

What users say, and what is missing

User reports are anecdotal, not clinical evidence. In Reddit posts counted by Dr. Pep, about 9% of 70 first-hand CJC-1295 reports mentioned better sleep.[18]

For ipamorelin, about 17% of 86 first-hand reports mentioned better sleep. About 5% mentioned injection site reaction, and about 4% said they stopped because of side effects.[18]

Several gaps are the same for both:

  • No controlled human study of non-DAC CJC-1295 exists, according to a 2026 review.[1]
  • No human trial in our records tested ipamorelin for muscle gain, fat loss, sleep or anti-aging.[10][3]
  • Our sources don't cover drug interactions for either one.[4][3]

The bottom line

The two peptides are not interchangeable, and the evidence for each is small. CJC-1295 DAC raised GH and IGF-1 for days in healthy adults, but that was a hormone reading, not a proven benefit.[2][4]

Ipamorelin released GH in a short dose study, and its one published efficacy trial showed no significant benefit.[10][3]

FDA's advisory committee voted against adding either to the compounding list. Long-term human safety is unknown for CJC-1295.[15][16][4]

This is a summary of what studies and regulators say, not advice.
